It will be an all-Ford front row for the 1000 kilometres classic with Ford Tickford Racing's Glenn Seton to start alongside the rookie sensation.
Ambrose was 11th after yesterday's qualifying session - but jumped up the order in the top-15 shootout.
"This is a credit to the team - they gave me a great car to go out and do the job today," said Ambrose.
"To come to Mount Panorama and grab pole in my first attempt is a dream come true - now my sights are set on the race.
"I'm under no illusions this is going to be tough race - but with the team on a high we are ready to give it our best shot.
Ambrose will team with Wayne Wakefield who earlier in the day won the Konica V8 race.
Even though Glenn Seton had the fastest time going into the shootout, he was still pleased to be on the front row for tomorrow's race.
"I'm delighted to be on the front row - and I feel very confident about our chances tomorrow," said Seton.
"This is definitely my best chance of winning this race.
"Myself, Steve Richards and the entire team are focused on giving FTR a win at the biggest race of the year.
The 161-lap V8 Supercar 1000 gets underway at 10am tomorrow morning.
