Mark Skaife scored his third straight overall win of the year, giving him a sizeable lead in the championship.
Ambrose earned runner-up with second place finishes in races one and two, with his weekend concluding with fourth place in the final 150-kilometre race.
"It was really important this weekend to be fast and consistent and that's what we've done," said Ambrose.
"This result further underlines that we're out to challenge our rivals and I know that we can improve even more.
"From a personal note I really enjoyed the hard racing this weekend. We pushed really hard throughout each of the races and the Falcon certainly stood up to the test.
"I'm now looking forward to Darwin with the aim of repeating our round win of last year in the top end."
Craig Lowndes returned to form at Eastern Creek, charging through the field in each of the three races, with the highlight being a super start in the final race.
The 00 Motorsport driver went from third position on the grid to first place by turn one. He held that lead up until just a hand full of laps before the end when he ran wide and dropped to second.
"It was important to gain a strong result this weekend, and the reward of third overall has us heading in the right direction," said Lowndes.
"We just missed the mark in qualifying - we'll improve in that area as we've shown our race pace is amongst the very best.
"The races were very hard on the driver as well as the car and tyres. The final laps of the last race were especially tough, but you have to give 100 percent in this sport if you want to gain results."
Ambrose has climbed to fourth place in the championship chase with Lowndes one place further back.
Ford next goes racing at Darwin's Hidden Valley Raceway from 17 - 19 May.
